今天算是好天。

1.
first thing to lab was to check my yeast....
heat-shock 比之前的 efficiency 高很多，
may have at least double amount of colonies...
gonna do 2 transformations this week...

2.
check for +ve clones on -Ade/-His plate...
5 of 7 grew...not too bad...
set up for auto-activation test and a/b-gal test...
Munner 等不急想知道是啥，已經準備好明天就送 sequencing...

3.
developing film for myristoylation...
got band for +ve control finally...
but, how come there's also a band for -ve control...?
anyways, we now know that BBS3 is not myristoylated...

4.
last thing...今天 lab 充滿了恭喜聲...
詳情沒聽清楚是啥，不過大概是 Oliver 求婚成功之類的吧....
they decide that wedding will be in next summer...
